Screening of Microorganism Producing Cellulase from Doenjang, a Korean Traditional Fermented Food and Characterization of Cellulase from Cellulolytic Microorganism

Abstract
Bacillus subtilis FB1 was isolated from Doenjang, a Korean traditional fermented food as a cellulolytic microorganism due to its ability of forming active zone on LB agar plate containing carboxymethylcellulose (CM-cellulose) as substrate with trypan blue. In the analysis of 16S rRNA gene sequence from the strain, the isolated strain showed 99% of identity with Bacillus subtilis, therefore we named the strain Bacillus subtilis FB1. The cellulase from B. subtilis FB1 exhibited the highest activities at pH 5.0 and 40¡ÆC conditions. And the cellulase from B. subtilis FB1 for the various cellulosic substrates such as CM-cellulose, alkali swollen cellulose, and Avicel has the highest activity for CM-cellulose. When the degradation products of alkali swollen cellulose by cellulase from B. subtilis FB1 were analyzed by thin-layer chromatography, the various size cellooligo-saccharides were detected as main products. From these results of enzyme activity for cellulosic substrates and the main enzyme reaction products, we concluded that the enzyme form B. subtilis FB1is one of the endo-type cellu.
